Lorsque la fonction mitochondriale est alt\u00e9r\u00e9e ou que le microenvironnement tumoral devient hypoxique, le NADH s&rsquo;accumule \u00e0 l&rsquo;int\u00e9rieur des cellules, perturbant cet \u00e9quilibre et cr\u00e9ant un stress m\u00e9tabolique. Les cellules normales ne peuvent g\u00e9n\u00e9ralement pas soutenir cet \u00e9tat et meurent. Les cellules canc\u00e9reuses, en revanche, peuvent r\u00e9orienter leurs voies m\u00e9taboliques pour \u00e9liminer en continu l&rsquo;exc\u00e8s de NADH, maintenant leur survie dans des conditions hostiles et d\u00e9veloppant m\u00eame une r\u00e9sistance aux m\u00e9dicaments. La voie m\u00e9tabolique compl\u00e8te sous-jacente \u00e0 ce m\u00e9canisme de survie n&rsquo;avait pas \u00e9t\u00e9 enti\u00e8rement \u00e9lucid\u00e9e jusqu&rsquo;\u00e0 pr\u00e9sent.    <\/strong><\/p>\n\n<figure class=\"wp-block-image size-full\"><img decoding=\"async\" width=\"685\" height=\"385\" src=\"https:\/\/www.celfull.com\/wp-content\/uploads\/2026\/08\/image-2.jpg\" alt=\"\" class=\"wp-image-28117\" title=\"\" srcset=\"https:\/\/www.celfull.com\/wp-content\/uploads\/2026\/08\/image-2.jpg 685w, https:\/\/www.celfull.com\/wp-content\/uploads\/2026\/08\/image-2-300x169.jpg 300w\" sizes=\"(max-width: 685px) 100vw, 685px\" \/><\/figure>\n\n<p class=\"wp-block-paragraph\"><strong>Cette \u00e9tude confirme que les cellules canc\u00e9reuses poss\u00e8dent une voie m\u00e9tabolique collaborative s&rsquo;\u00e9tendant des mitochondries au cytoplasme. La pyruvate carboxylase (PC) sert de point de d\u00e9part, agissant de concert avec la malate d\u00e9shydrog\u00e9nase et l&rsquo;enzyme malique 1 (ME1) pour former un efficace \u00ab\u00a0syst\u00e8me tampon du NADH\u00a0\u00bb. Ce syst\u00e8me comprend deux voies parall\u00e8les : l&rsquo;une consomme le NADH accumul\u00e9 dans les mitochondries, l&rsquo;autre \u00e9limine l&rsquo;exc\u00e8s de NADH dans le cytoplasme. Les deux voies convertissent en fin de compte les \u00e9quivalents r\u00e9ducteurs exc\u00e9dentaires en NADPH, soulageant simultan\u00e9ment la pression m\u00e9tabolique de l&rsquo;accumulation de NADH et reconstituant les mat\u00e9riaux n\u00e9cessaires \u00e0 la d\u00e9fense antioxydante cellulaire, favorisant ainsi la survie des cellules canc\u00e9reuses dans des conditions de stress.   <\/strong><\/p>\n\n<figure class=\"wp-block-image size-full\"><img decoding=\"async\" width=\"645\" height=\"332\" src=\"https:\/\/www.celfull.com\/wp-content\/uploads\/2026\/08\/image-1.jpg\" alt=\"\" class=\"wp-image-28116\" title=\"\" srcset=\"https:\/\/www.celfull.com\/wp-content\/uploads\/2026\/08\/image-1.jpg 645w, https:\/\/www.celfull.com\/wp-content\/uploads\/2026\/08\/image-1-300x154.jpg 300w\" sizes=\"(max-width: 645px) 100vw, 645px\" \/><\/figure>\n\n<p class=\"wp-block-paragraph\">(Source: The Innovation, 2026)<\/p>\n\n<p class=\"wp-block-paragraph\"><strong>L&rsquo;\u00e9tude a \u00e9galement produit un r\u00e9sultat qui remet en cause les connaissances existantes. L&rsquo;enzyme m\u00e9tabolique G6PD (glucose-6-phosphate d\u00e9shydrog\u00e9nase), largement reconnue pour son r\u00f4le dans la g\u00e9n\u00e9ration de NADPH par sa propre r\u00e9action catalytique, s&rsquo;est r\u00e9v\u00e9l\u00e9e remplir une fonction allant au-del\u00e0 de son r\u00f4le traditionnel dans cette voie. La recherche d\u00e9montre que la G6PD peut agir comme un \u00ab\u00a0\u00e9chafaud mol\u00e9culaire\u00a0\u00bb, se liant directement \u00e0 la ME1 et l&rsquo;activant, acc\u00e9l\u00e9rant ainsi l&rsquo;ensemble de la voie de conversion du NADH. Cette fonction d&rsquo;\u00e9chafaudage op\u00e8re ind\u00e9pendamment de l&rsquo;activit\u00e9 catalytique propre de la G6PD.   <\/strong><\/p>\n\n<p class=\"wp-block-paragraph\"><strong>D&rsquo;une importance translationnelle particuli\u00e8re, l&rsquo;\u00e9quipe de recherche a valid\u00e9 l&rsquo;efficacit\u00e9 antitumorale du ciblage de cette voie. Lorsque le compos\u00e9 de petite mol\u00e9cule ZY444 a \u00e9t\u00e9 utilis\u00e9 pour bloquer la PC, un n\u0153ud cl\u00e9 de la voie, la capacit\u00e9 tampon du NADH des cellules canc\u00e9reuses a \u00e9t\u00e9 consid\u00e9rablement affaiblie. Ceci a sensibilis\u00e9 les cellules \u00e0 la metformine, un inhibiteur de la respiration mitochondriale, et au CB839, un inhibiteur du m\u00e9tabolisme de la glutamine.
